Owkin and BeOne Medicines Sign Clinical Supply Agreement for Tislelizumab in INVOKE Trial of OKN4395

Paris - June 1, 2026 - Owkin, the Agentic AI company pioneering Biological Artificial Superintelligence to revolutionize drug discovery and development, today announced a clinical supply agreement with BeOne Medicines. Under the agreement, BeOne will supply tislelizumab, a PD-1 inhibitor, for use in combination with OKN4395 in Owkin's ongoing INVOKE Phase 1a/1b trial in patients with advanced solid tumors.
OKN4395 is a first-in-class small-molecule triple antagonist of EP2, EP4, and DP1 - three immunosuppressive receptors downstream of the COX-2 pathway. Specifically, our AI-scientist Owkin K Pro uncovered the PGD2-DP1 axis as a novel immunosuppressive mechanism, which was fully validated in the lab. Our internal data revealed a hidden context dependent mechanism in which the well-known highly potent anti-tumor function of EP2/EP4 dual blockade was fully inhibited by PGD2-mediated DP1 signaling, highlighting the crucial need for triple EP2, EP4 and DP1 blockade in those tumors.

Response to checkpoint inhibitors alone remains suboptimal in some tumors, particularly those that are immunologically 'cold'. Blocking PGE2 and PGD2 immunosuppressive pathways in combination with checkpoint inhibitors could sensitize tumors that respond poorly to checkpoint inhibitors.
